Is the value or desirability any higher for an early production (1954 DOM, serial # 32xx) Ruger Single Six - flat gate the single action revolver? Condition is about 90-95%.

Oh, that's something I've never seen before. I'm definitely wrong about my value estimates below in my original reply.
The value as described is at least $300 to maybe as much as twice that to a serious collector that wants that particular model.
Current auctions for 3 screw single six have starting prices of $350 to $499 but only 2 guns among those have bids.
Edit = if this has been converted to new model configuration $300 would be about top dollar.
Edit #2 = I'm not sure exactly what "flat gate" means, therefore my figures above may be low.0 -
The "flat gate" s-6,s in good condition are good collector guns. The conversion doesn't hurt the value as Ruger returns the original parts to you [;)] 0 -

The conversion "shouldn't hurt value", since the last I knew, yes, Ruger returns the original parts,
That said I have encountered a discouraging number of 3 screws for which the original parts have been lost somewhere along the way. If that's the case, finding replacements can be very hard to find and costly to replace. I have no problem with the New Model mechanism in New Model guns. They can be very smooth, even if they lack the requisite clicks that we all love. The converted revolvers I've handled however have been rough, butt ugly and deal killers down to the last one. Buyer beware.0
