I'd be looking at the bolt and the hand (#4 and 10) with their spring.

So will it function correctly when unloaded and binds after the first shot. The balls are staying seated and the spent cap is clear.

added: The hand and bolt are fitted pieces, perhaps they were replace without being fitted. Nipples could be short, the cylinder could be wrong. You need to get some hands on help from a gunsmith or other qualified person.

Okay here is what I noticed. When the gun is apart and I move the cylinder forward just a little bit, it will function properly but the hammer will not strike the cap. When I put it all back together and everything looks like it is alined, the cylinder will not rotate. Looks like it is the wrong cylinder[?] I don't know and am guessing.

Probably too tight a fit of barrel to cylinder. After firing there is fouling there that causes binding. The Colts were hand fitted to allow for this problem and still able to fire the percussion cap. On reproductions this can be a reliability issue. After loosening the wedge a little you may need longer nipples.

On my Series 3 Colt 1860 two things will cause similar binding 1, pushing the barrel locking wedge too far into the slot - this appears to push the cylinder back against the recoil shield and bind everything up; 2, small pieces of the fired cap get into the hand slot and bind everything tight, but fall clear as soon as the cylinder is pulled forward and all pressure is released off the hand.

WARNING! do NOT use longer nipples unless a qualified gunsmith looks at it! I did that on an original colt I rebuilt, BECAUSE THE HAMMER WASN'T STRIKING THE CAPS, and the recoil slammed the cylinder back against the frame, firing all six chambers due to the long nipples striking the frame. One ball went down the barrel, four cleared the frame, and the 6th was jammed in the loading lever. Recoil damn near broke my wrist.

    The hand is too long.
It's a careful file and try proposition.
